Neural Stem Cell Expansion Medium
Neural Stem Cell Expansion Medium (NSCEM) consists of 100 ml of basal medium and 2 ml of Neural Stem Cell Expansion Growth Supplement (NSCEGS, Cat #5992). Note: NSCEGS is not pre-mixed in NSCEM; it must be added separately to make the complete NSCEM.
Neural Stem Cell Expansion Medium (NSCEM), when used with Neural Stem Cell Expansion Growth Supplement (NSCEGS, Cat #5992) is a complete medium designed for optimal growth of mammalian neural stem cells in vitro. It is a sterile, liquid medium which contains essential and non-essential amino acids, vitamins, organic and inorganic compounds, hormones, growth factors and trace minerals. The medium is HEPES and bicarbonate buffered and has a pH of 7.4 when equilibrated in an incubator with an atmosphere of 5% CO 2 /95% air. The medium is formulated (quantitatively and qualitatively) to provide an optimally balanced nutritional environment that selectively supports the growth of mammalian neural stem cells in vitro.

Figure 1. Expansion of Neural Stem Cells in Neural Stem Cell Expansion Medium
NSC cultured in Neural Stem Cell Expansion Medium (NSCEM) could be stably expanded to generate large number of cells. 1:3-1:6 passaging can be achieved upon each passage. NSCs (Cat#1650, #1660) were derived using HPSC Neural Induction Medium (Cat#5931). For passaging and banking, Neural Stem Cells were dissociated using CellEase (Cat#5813) and banked with StemCryo (Cat#0153)Ā

Figure 2. Neural Stem Cells Cultured in Neural Stem Cell Expansion Medium Show Minimal Spontaneous Neuronal Differentiation
Passages 1 (A) and 5 (B) of a representative NSC culture maintained in Neural Stem Cell Expansion Medium. Cells were immunolabeled with Nestin (green) to identify NSCs (counter stained with DAPI in blue). Spontaneous neuronal differentiation is low in NSC cultures maintained in Neural Stem Cell Expansion Medium. A and B were taken at the same magnification.

Figure 3. Neural Stem Cells Maintained in Neural Stem Cell Expansion Medium can Differentiate into Neurons and Astrocytes
NPCs can differentiate into neurons (A, β-tubulin shown in green) with Neuronal Medium (Cat#1521) and astrocytes (B, GFAP shown in green) with AM (Cat#1801).
