ScienCell's GeneQuery™ Human Osteoarthritis and Cartilage Repair qPCR Array (GQH-OAC) facilitates gene expression profiling of 88 key genes involved in the establishment and progression of osteoarthritis (OA), focusing on the breakdown of cartilage repair. OA is characterized by joint swelling, cartilage degradation, and eventually bone breakdown. While the causes of OA are still unknown, there are genetic components that can determine a person’s susceptibility to the disease. Brief examples of how included genes may be grouped according to function are shown below:
- Cartilage homeostasis: FGF18, FGF2, HIF1A, IGF1, SMAD2, TGFB1
- Inflammation: IL17A, ELF3, IL18, MAPK3, PTGS2, TNF
- Chondrocyte hypertrophy: CCL19, SIRT1, WNT4, IHH, MMP13
- Susceptibility genes: SMAD3, ASPN, FRZB, DIO2
- Markers of moderate or severe OA: TIMP1, COL2A1, FN1, C11orf31, CILP
GeneQuery™ qPCR array kits are qPCR ready in a 96-well plate format, with each well containing one primer set that recognizes and efficiently amplifies a specific target gene's cDNA. The carefully designed primers ensure that: (i) the optimal annealing temperature in qPCR analysis is 65°C (with 2 mM Mg2+ and no DMSO); (ii) the primer set recognizes all known transcript variants of the target gene, unless otherwise noted; and (iii) only one gene is amplified. Each primer set has been validated by qPCR with melt curve analysis and gel electrophoresis.
