GeneQuery™ Human Huntington’s Disease qPCR Array Kit
ScienCell’s GeneQuery™ Human Huntington’s Disease qPCR Array Kit (GQH-HUN) profiles 88 key genes involved in the development and progression of Huntington’s disease.
ScienCell’s GeneQuery™ Human Huntington’s Disease qPCR Array Kit (GQH-HUN) profiles 88 key genes involved in the development and progression of Huntington’s disease. Huntington’s disease is an autosomal-dominant neurodegenerative disorder characterized by dystonia, cognitive decline, and difficulties with coordination and behavior. In this disease, an expanded CAG repeat leads to the production of a mutant form of the protein huntingtin (HTT) that has an abnormally long polyglutamine (polyQ) expansion. This polyQ expansion confers a toxic gain of function that makes the mutant huntingtin prone to aggregation and accumulation, resulting in neurodegeneration. Onset of symptoms typically occurs once an individual has reached middle-age; however, in individuals who possess a longer CAG repeat, the disorder has the potential to manifest earlier. Brief examples of how included genes may be grouped according to their functions are shown below:
- HD development/early pathogenesis: BAX, BDNF, CDK5, HTT, IGF1, MAPK1, NLRP3, PDE10A, SREBF2, TRIP10
- High Htt/mHtt interaction: CASP6, CREB1, DISC1, DNM1L, FOXP2, GRM5, HIP1, HIPK3, HMGB1, MAPK11, PRMT6, SLC12A5, SQSTM1, UBR5, ZDHHC17
- Associated with age at onset: ADORA2A, DRD2, FAN1, HAP1, LIG1, MAP2K6, MAP3K5, MC1R, MSH3, OGG1, PMS1, PMS2, PPARGC1A, SLC2A3, TCERG1, UCHL1
- Upregulated in HD: ADAM10, AKT1, ARFGEF2, ASIC1, CD40, CNR2, LGALS3, NPY, PARP1, PPIF, SIRT1, TRPC5, TWIST1
- Downregulated in HD: CNR1, CPLX2, DHFR, DRD1, DYNLT1, HSF1, JPH3, MFN1, MFN2, RAC1, UBE3A
- Neuroprotective effects: BAG1, CNTF, FGF9, GRIK4, GRM2, GRM3, IGF2, IRS2, PIAS1
- Involved in late-stage pathogenesis: BCL2, FIS1, OPA1, TOMM40
- Cholesterol metabolism: CYP46A1, SLC30A3
- Potential therapeutic target: GPR52, HDAC4, MYRF, NFE2L2, SIRT2
- Associated with CAG repeats/expansion: HDAC3, MLH1, MLH3
Note: all gene names follow their official symbols by the Human Genome Organization Gene Nomenclature Committee (HGNC).
GeneQuery™ qPCR array kits are qPCR ready in a 96-well plate format, with each well containing one primer set that can specifically recognize and efficiently amplify a target gene's cDNA. The carefully designed primers ensure that: (i) the optimal annealing temperature in qPCR analysis is 65°C (with 2 mM Mg2+, and no DMSO); (ii) the primer set recognizes all known transcript variants of target gene, unless otherwise indicated; and (iii) only one gene is amplified. Each primer set has been validated by qPCR with melt curve analysis, and gel electrophoresis.
