纤连蛋白细胞黏附检测

Cell adhesion plays an important role in cellular communication and regulation, and is of fundamental importance in the development and maintenance of tissues. Fibronectin is a high-molecular-weight extracellular matrix glycoprotein involved in many cellular processes, including tissue repair, embryogenesis, blood clotting, and cell migration/adhesion. The ScienCellTM Fibronectin Cell Adhesion Assay is designed for the rapid, quantitative and reliable measurement of cell adhesion to fibronectin. The kit is composed of a 48-well plate pre-coated with fibronectin, as well as BSA as negative controls (see plate format in Figure 1). Cells are cultured in the pre-coated wells for a desired period of time, then unbounded cells are washed away, and the adhered cells are fixed and stained, followed by an extraction step which leads to dye elution from stained cells into the supernatant. Thus cell adhesion can be quantified using a colorimetric ELISA plate reader at 595 nm.

本试剂盒提供一块预包被纤连蛋白的 48 孔板，并配有以 BSA（牛血清白蛋白）包被的阴性对照孔（孔板布局见图 1）。实验时，将细胞接种至预包被孔中培养至所需时间，随后洗去未黏附的细胞，对黏附细胞进行固定和染色，再通过染料提取步骤将染色细胞中的染料释放至上清液中。最后，使用酶联免疫吸附测定（ELISA）酶标仪在 595 nm 波长下进行比色检测，即可对细胞黏附水平进行定量分析。
